Reliability of Angiotensin-Converting Enzyme Gene Polymorphism in Testing

IN RESPONSE:
Odawara and Yamashita question the reliability of the methods we used for genotyping the insertion/deletion polymorphism of the ACE gene [1, 2]. It has been emphasized that using primers that flank the polymorphic region may result in overestimation of the DD genotype because of preferential amplification of the D allele [2, 3]. We therefore reassessed all our samples using the method proposed by Shanmugan and colleagues [2], as described in the Methods section of our report. We also used the method of Lindpaintner and associates [3] to retype the samples that were previously found to be DD homozygous; this method also uses an insertion-specific primer. This reassessment completely confirmed the data reported in our study. Therefore, we are confident that mistyping did not jeopardize the validity of our data.
